Abstract

ABSTRACT A method (100) for characterizing bacteriophages comprising the steps of collecting sewage water samples from a natural source, processing the sewage samples with a phage buffer solution to obtain viable and concentrated solution of bacteriophages, isolating the bacteriophages from the concentrated solution of bacteriophages by performing a double-layer agar plaque assay, determining the bacteriophage-induced plaque formations based on the double-layer agar plaque assay and characterising genetic material of bacteriophages of the bacteriophage-induced plaque formations.

[0001] This invention generally relates to a field bacteriophages and in particular relates to a method for characterising bacteriophages.
BACKGROUND

[0003] The rapid rise of multi-drug resistant (MDR) bacterial infections has posed significant challenges in treating common bacterial diseases, resulting in increased mortality, prolonged hospital stays, and substantial healthcare costs. Traditional antibiotic treatments are increasingly ineffective against M , Claims:1. A method (100) for characterizing bacteriophages, the method (100) comprising the steps of:
collecting sewage water samples from a natural source;
processing the sewage samples with a phage buffer solution to obtain viable and concentrated solution of bacteriophages;
isolating the bacteriophages from the concentrated solution of bacteriophages by performing a double-layer agar plaque assay;
determining the bacteriophage-induced plaque formations based on the double-layer agar plaque assay; and
characterising genetic material of bacteriophages of the bacteriophage-induced plaque formations.

2. The method (100) as claimed in claim 1, wherein the bacteriophage-induced plaque formations exhibit lytic activity against target multi-drug resistant bacteria, including at least Staphylococcus aureus and Pseudomonas aeruginosa.
