Abstract

Abstract A unique technique for the isolation and molecular identification of Synechococcus elongatus from a freshwater body is the subject in the current invention. This innovation involves in the isolation of cyanobacterium specifically by combining cutting-edge microbiological methods with the ideal growth conditions. 16S rRNA gene sequencing is then used to identity and describe the isolated cynobacterium, enabling accurate phylogenetic categorization and distinction from previously recognized bacteria.The isolated strain's genetic variation, physiological, and metabolic characteristics were also discovered, which can be advantageous in several biotechnological applications. These also includes synthesizing high-value bioactive chemicals, carbon sequestration, environmental bioremediation, and the production of biofuels. The defined strain and the unique separation method have the potential to improve the efficiency of other microbially driven biotechnological breakthroughs and photosynthesis activities. This innovation is also advances our understanding of the diversity of microorganisms found in freshwater environments and ~hells light on the function of cyanobacteria in the cycling of nutrients and ecological equilibrium. The recently identified strain has a great potential for environmentally and industrially usage, especially when combined with the newly established techniques for its separation and identification.

The current invention is performed on the separation, identification, and characterisation of microbial species from natural its habitats and falls under the fields of molecular biology,
biotechnology, and microbiology. The invention specifically involves to the use of cuttingedge
molecular and microbiological techniques to isolate a novel strain of the
cyanobacterium Synechococcus elongatus from a freshwater body. It inCludes the ideal
isolation procedures, the use of 16S rRNA gene sequencmg for accurate phylogenetic
identification, and the establishment of selective culture conditions.
This invention is also discusses on the novel strain of Synechococcus elongatus's umque
physiological, metabolic, and genetic characteristics, as well as its ecological significance in
freshwater habitats and prospective industrial uses. These strain's applications is used in
many process, include the production of bio-diesel, photosynthetic carbon fixation.
Furthermore, the invention investigates the strain's possible applications in sustainable
biotechnological processes, including carbon sequestration, bio-based product manufacture,
and optimization of bio process efficiency in certain environmental or artificial conditions.
With potential uses in freshwater management and conservation, the idea also has
significance for ecological studies, contributing to understanding the microbial diversity and
the role of cyanobacteria in aquatic environments

Claims
I. The invention discloses the isolation of Synechococcus e/ongatus bacteria from the
fresh water bodies. Here the fresh water is collected from the wells that are used for
the agriculture land, which are completely exposed to the sun for a long time.
2. As indicated in claim I, to isolate Synechococcus elongatus, the BG - I I media
should be prepared by adding NaNO, (Sodium nitrate), KzHPO. (Dipotassium
phosphate), MgS0.7HzO (Magnesium sulfate), CaCb2HzO (Calcium chloride),
Citric acid, Ferric ammonium citrate, EDTA (Disodium salt), NazCO, (Sodium
carbonate) and essential materials in trace element in IL distilled water and adjust it
to 7 to 7.5 pH. Then autoclave the prepared medium, and store it.
]. As indicated in claim 2, the prepared the BG - I I agar media was poured into the
petri dishes and innoculate the collected fresh water sample after serial dilution on it
and spread it using the sterile L - rod and then incubate it 25 c to 30" C under the light
cycle upto 24-48 hours.
4. As indicated in claim 3, the isolated bacterium Synechococcus elongatus must need to
be characterized to check the genomic DNA of the species. The 16s rRNA sequencing
to analyse the gene sequence of the isolated microbial culture by Sanger sequencing.
5. As indicated in claim 4, the gene sequence analyzed for the identification of the
specific gene which are responsible for the production of the high quality Pectinase
enzyme.
