Specification

Description:FIELD OF THE INVENTION
This invention relates to agricultural and biotechnological applications, specifically to a nutrient-enriched medium using Cloeme viscosa (CVE) designed to optimize the growth, sporulation, and metabolite production of the biocontrol agent Trichoderma viride. The CVE medium enhances the efficacy of T. viride, supporting its application in biocontrol, agriculture, and environmental biotechnology.
BACKGROUND OF THE INVENTION
Trichoderma viride is a fungal species widely used as a biological control agent in agriculture and environmental biotechnology due to its ability to suppress plant pathogens and promote plant growth. However, conventional media for T. viride cultivation, such as Potato Dextrose Agar (PDA) and Sabouraud Dextrose Agar (SDA), are not specifically tailored to maximize the growth and biocontrol efficacy of T. viride. These standard media lack essential nutrients and conditions that support optimal growth, sporulation, and production of secondary metabolites, which are critical for its effectiveness in biocontrol applications. Consequently, the limited growth and activity of T. viride on these media restrict its potential impact and efficiency in field applications.
Current research highlights the importance of tailored nutrient profiles that cater to the unique metabolic needs of T. viride. Studies show that specific nitrogen and carbon sources, as well as trace minerals, significantly influence the growth and efficacy of T. viride as a biocontrol agent. However, existing formulations lack the specific nutrient enrichment necessary to maximize these attributes. This invention addresses this gap by introducing an enriched medium formulated with Cloeme viscosa extracts, which provide essential nutrients and growth factors, leading to enhanced biomass production, sporulation, and metabolite synthesis in T. viride. The CVE medium thus represents a novel approach to improving the efficacy of T. viride in various agricultural and environmental applications.
SUMMARY OF THE INVENTION
This summary is provided to introduce a selection of concepts, in a simplified format, that are further described in the detailed description of the invention.

The Cloeme Viscosa Enriched (CVE) Medium for Trichoderma viride is an innovative nutrient formulation that enhances the growth, sporulation, and metabolite production of T. viride. The medium preparation begins with the extraction of bioactive compounds from fresh or dried Cloeme viscosa leaves, which are known for their nutrient richness, including proteins, carbohydrates, and phytohormones that promote fungal growth. The extraction process involves washing the plant material, followed by boiling or soaking in distilled water to release the bioactive components. The resulting extract is filtered to remove solid residues, retaining a nutrient-rich liquid phase used in the CVE medium.
The enriched medium combines Cloeme viscosa extract with nitrogen sources like ammonium sulfate or urea, which are critical for protein synthesis and robust fungal growth. Yeast extract is added to supply B-vitamins and amino acids, supporting metabolic activity and enhancing sporulation. Carbon sources such as glucose and starch provide both immediate and sustained energy for T. viride, facilitating balanced growth and activity. Mineral supplements, including calcium carbonate and magnesium sulfate, are incorporated to maintain pH stability and provide essential minerals. Additionally, trace elements like iron, zinc, and manganese are added in chelated forms, supporting enzymatic functions crucial for fungal metabolism.
To further optimize growth, vitamins and growth factors, including thiamine and biotin, are included in the medium to enhance metabolic pathways. The incorporation of hormones, such as auxins and gibberellins derived from Cloeme viscosa, promotes hyphal extension and sporulation, significantly improving the overall biomass yield. Agar-agar may be added as a gelling agent for solid media formulations, enabling plate assays and other laboratory applications. The prepared CVE medium is sterilized through autoclaving and adjusted to a pH of 6.0 to 6.5 for optimal T. viride growth conditions. After cooling, the medium is inoculated with T. viride under sterile conditions, followed by incubation at controlled temperatures to assess mycelial density, sporulation, and secondary metabolite production.
